目的:探讨丙泊酚对幼鼠认知以及神经细胞中Caspase-3表达的影响。方法:选取15~18日龄的SD幼鼠48只,随机分为A、B、C、D四组各12只。A组注射生理盐水(10 mL/kg),B、C、D三组分别注射丙泊酚50、100、200 mg/kg。连续注射6 d,前5 d做水迷宫实验检测幼鼠学习能力,第6天撤去平台检测幼鼠记忆能力。6 d后处死幼鼠取脑组织,分别采用免疫组化与Western Blotting检测Caspase-3的表达情况。结果:与A组、B组比较,C组、D组的平台潜伏期长、上台前路程长、穿越平台次数少、上台前时间长、Caspase-3表达量高,差异均有统计学意义(P均<0.05)。结论:高剂量(100~200 mg/kg)的丙泊酚可降低幼鼠的空间学习能力和记忆能力,提高神经细胞中Caspase-3的表达。 |

Effect of Propofol on Cognitive Function and the Expression of Caspase-3 in Immature Rat |

Abstract: |
Objective: To discuss the effects of propofol on cognitive function and the expression of caspase-3 in immature rats. Methods: Fifteen to eighteen days old of forty-eight immature SD rats were selected, and divided into 4 groups randomly (12 each), group A injected NS 10 mL/kg, group B injected propofol 50 mg/kg, group C injected propofol 100 mg/kg and Group D injected propofol 200 mg/kg for six days consecutively. The tests of learning ability were performed in Morris water maze everyday at the initial five days, and the tests of memory ability were performed in Morris water maze at the 6th day. The rats were sacrificed on the 6th day after the Morris water maze test to obtain the brain specimen, and the expression of caspase-3 was detected by immunohistochemistry and Western Blotting. Results: The latency period, time and distance before mounting platform in group C and group D were significantly longer than those in group A and group B (P<0.05) and frequency of crossing platform in group C and group D were significantly fewer than that in group A and group B (P<0.05). The expression of caspase-3 in group C and group D were significantly higher than that in group A and group B (P<0.05). Conclusion: Propofol 100 mg/kg and 200 mg/kg can decrease spatial learning and memory in immature rat and increase the expression of caspase-3. |
