高效液相色谱法测定新生儿体内头孢他啶的血药浓度

摘要:
目的:建立基于少量血样测定新生儿体内头孢他啶血药浓度的方法,研究头孢他啶在新生儿体内代谢特点。方法:采用高效液相色谱(HPLC)法,Diamonsil C18色谱柱(250 mm×4.6 mm, 5 µm),以甲醇-0.02 mmol/L磷酸二氢钾溶液(15:85)为流动相,流速1 mL/min,柱温30 ℃,检测波长254 nm;采用饱和硫酸铵溶液沉淀蛋白后进样。结果:头孢他啶浓度在1.0-200 µg/mL 范围内与头孢他啶峰面积/内标峰面积呈良好的线性关系(r=0.999 9),低、中、高3个浓度的方法回收率为99.54%-100.13%,RSD<8%;提取回收率为91.06%-94.58%,RSD<5%;头孢他啶在新生儿体内代谢个体差异较大。结论:本方法采血量少,操作简便准确,回收率稳定,适用于头孢他啶在新生儿体内的药动学研究。

Determination of Blood Concentration of Ceftazidime in Neonates by HPLC

Abstract:
Objective: To establish a method for determination of blood concentration of ceftazidime in neonates based on a small amount of blood samples, and to study the metabolic characteristics of ceftazidime in neonates. Methods: The HPLC separation was performed on Diamonsil C18 column (250 mm×4.6 mm, 5 µm). The mobile phase was methamo-0.02 mmol/L potassium dihydrogen phosphate (15:85) at the flow rate of 1 mL/min. The column temperature was 30 ℃, with detection wavelength of 254 nm. The sample was deposited by saturated ammonium sulfate solution and then injected. Results: There was a good linear relationship between the ratio of ceftazidime peak area and the internal standard peak area within the range of 1.0-200 µg/ mL (r =0. 999 9). The recovery rates of low, medium and high concentrations were 99.54%-100.13%, RSD<8%; the extraction recovery was 91.06%-94.58%, RSD<5%; and ceftazidime was significantly different in neonatal metabolism. Conclusion: The method has the advantages of small blood collection, simple and accurate operation, stable recovery rate and is suitable for pharmacokinetic study of ceftazidime in neonates.
