小儿化滞健脾丸对幼龄厌食模型大鼠胃窦和外周血胃动素及微量元素的影响

摘要:
目的:建立幼龄大鼠厌食症模型,评价小儿化滞健脾丸的治疗作用并探讨其对大鼠胃窦组织和外周血中胃动素(MTL)及微量元素锌、铁、铜等的影响。方法:将72只SD幼龄大鼠随机分为空白对照组、模型对照组、阳性对照组及小儿化滞健脾丸大、中、小剂量组六组各12只,用特制饲料喂养法构建厌食症模型,给药4周,监测体质量和摄食量等一般状态。末次完成给药后,测定胃内残留率;利用火焰原子吸收法检测血浆锌、铁、铜含量;采用ELISA试剂盒检测血浆中MTL的含量;采用免疫组化法检测胃窦组织中MTL的表达情况。结果:与空白对照组比较,模型对照组大鼠的摄食量、体质量、胃窦MTL表达及外周血锌、铁、MTL含量均降低,胃内残留率增高(P均<0.05)。与模型对照组比较,各治疗组大鼠的摄食量、体质量、MTL、铁含量均增高(P均<0.05);阳性对照组、大剂量组锌含量增高(P均<0.05);阳性对照组、大剂量组、中剂量组胃内残留率降低(P均<0.05)。结论:小儿化滞健脾丸可以有效改善幼龄大鼠的厌食症状,作用机制可能为通过调节外周血微量元素水平和MTL的分泌及胃窦组织MTL的表达,即调节“脑肠肽-食欲中枢”来发挥其治疗作用。

Abstract:
Objective: To establish the anorexia model in immature rats, evaluate the therapeutic effects of Xiao’er Huazhi Jianpi pills, and explore its effects on motilin (MTL) and trace elements such as Zn, Fe and Cu in gastric antrum tissue and peripheral blood of rats. Methods: Totally 72 SD immature rats were randomly divided into the blank group, the model group, the positive group, and Xiao’er Huazhi Jianpi pills high, medium and low dose group, with 12 rats in each group. The anorexia model was established by special feeding method, with administration for 4 weeks, general conditions such as body mass and food intake were monitored. After the last administration, the gastric residual rate was determined. The contents of Zn, Fe and Cu in plasma were detected by flame atomic absorption spectrometry. The contents of MTL in plasma were detected by enzyme-linked immunosorbent assay (ELISA). Immunohistochemistry was used to detect the expression of MTL in gastric antrum tissues. Results: Compared with the blank group, the food intake, body mass, expression of MTL in gastric antrum tissues and contents of MTL in the model group decreased significantly (P<0.05), while gastric residual rate increased significantly (P<0.01). Compared with the model group, the food intake, body mass, MTL, content of Fe of the rats in each treatment group increased significantly (P<0.05), the content of Zn in the positive group and Xiao’er Huazhi Jianpi pills high dose group increased significantly (P<0.01), the residual rate in the positive group, Xiaoer Huazhi Jianpi Pills high, medium dose group decreased significantly (P<0.05). Conclusion: Xiao’er Huazhi Jianpi pills can effectively improve the anorexia symptoms in the immature rats. The mechanism may be through regulating the levels of trace elements in the peripheral blood, the secretion of MTL and the expression of MTL in gastric antrum tissue, namely, regulating the “brain-intestinal peptide-appetite center” to achieve its therapeutic effects.
