小儿鼻炎片对幼龄动物慢性鼻炎的治疗作用及机制初探

摘要:
目的:观察小儿鼻炎片对幼龄大鼠二氧化硫染毒致慢性鼻炎的保护作用,并对其抗炎、免疫作用进行初步探索。方法:将幼龄大鼠随机分为空白对照组、模型组、辛夷鼻炎丸组、小儿鼻炎片高剂量组、小儿鼻炎片中剂量组、小儿鼻炎片低剂量组。将幼龄小鼠随机分为模型组、辛夷鼻炎组及小儿鼻炎片高、中、低剂量组。除空白对照组外,其余大鼠均采用二氧化硫熏蒸法建立幼龄大鼠慢性鼻炎模型,各给药组给予相应药物灌胃,连续4 周后,观察小儿鼻炎片对慢性鼻炎大鼠模型鼻黏膜病理组织学改变、幼龄小鼠碳粒廓清率、幼龄大鼠棉球肉芽肿形成的影响。结果:幼龄大鼠经二氧化硫熏蒸后,模型组大鼠鼻黏膜上皮出现变性,血管扩张充血,黏膜层病理损伤较重。给药后,小儿鼻炎片高剂量组(1.32 g/kg) 和中剂量组(0.66 g/kg)大鼠鼻腔黏膜的病理损伤程度较模型组明显减轻,病理等级评分与模型组比较差异有统计学意义(P均<0.05);小儿鼻炎片高 (1.98 g/kg)、中(0.99 g/kg)、低(0.50 g/kg)剂量组小鼠的碳粒廓清指数高于模型组,差异有统计学意义(P<0.05);小儿鼻炎片高(1.32 g/kg)、中(0.66 g/kg)、低(0.33 g/kg) 剂量组大鼠的棉球肉芽肿干重与模型组比较降低,且差异有统计学意义(P<0.05)。结论:小儿鼻炎片可显著改善幼龄大鼠二氧化硫染毒后所引起的慢性鼻炎,对鼻黏膜有一定的保护作用,同时可不同程度地抑制大鼠棉球肉芽肿的形成,增强小鼠网状内皮吞噬系统的防御能力,有一定的免疫保护作用,这可能是其治疗慢性鼻炎的作用机制之一。

Abstract:
Objective: To observe the protective effects of Xiao’er Biyan tablets on chronic rhinitis induced by sulfur dioxide in young rats, and to preliminarily explore its anti-inflammatory and immune effects. Methods: The young rats were randomly divided into the blank control group, model group, Xinyi Biyan pills group, Xiao’er Biyan tablets high-dose, medium-dose and low-dose group. The young mice were randomly divided into the model group, Xinyi Biyan pills group, Xiao’er Biyan tablets high-dose, medium-dose and low-dose group. In addition to the blank control group, chronic rhinitis model of young rats was established by sulfur dioxide fumigation in all rats. Each dose group was given drug lavage. After 4 weeks, the effects of Xiao’er Biyan tablets on the pathological changes of nasal mucosa, carbon clearance rate and granuloma formation in young rats with chronic rhinitis were observed. Results: After being fumigated by sulfur dioxide, the mucosal epithelium of rats in the model group showed degeneration, vasodilatation and hyperemia, and the pathological damage of mucosa layer was severe. After administration, the degree of pathological damage of the nasal mucosa in Xiao’er Biyan tablets high-dose (1.32 g/kg) and medium-dose (0.66 g/kg) group decreased significantly compared with the model group, and the pathological grade was significantly different from the model group (P<0.05). The rate of carbon granular clearance in the mice with high-dose (1.98 g/kg), medium-dose (0.99 g/kg) and low-dose (0.50 g/kg) Xiao’er Biyan tablets was significantly higher than that of the model group, and the differences were statistically significant (P<0.05). Compared with the model group, the dry weight of cotton granuloma in Xiao’er Biyan tablets high-dose (1.32 g/kg), medium-dose (0.66 g/kg) and low-dose group (0.33 g/kg) decreased significantly, and the differences were statistically significant (P<0.05). Conclusion: Xiao’er Biyan tablets can significantly improve the chronic rhinitis induced by sulfur dioxide in young rats, and has a certain protective effect on the nasal mucosa. Meanwhile, it can inhibit the formation of cotton ball granuloma in rats to varying degrees and strengthen the reticuloendothelial in mice. The defensive capacity of the phagocytic system has a certain immune protection effect, which may be one of the mechanisms of treatment of chronic rhinitis.
