新疆地区疑似支气管哮喘学龄前患儿低亲和力IgE受体、白三烯C4合成酶基因多态性分析

摘要:
目的:探讨低亲和力IgE受体(FCER2)基因T2206C位点、白三烯C4合成酶(LTC4S)基因A444C位点基因型和等位基因在新疆哈萨克族、汉族、维吾尔族疑似气管哮喘学龄前患儿中的差异性分布特征,为新疆不同民族支气管哮喘学龄前患儿提供哮喘早期诊断参考。方法:采用荧光染色原位杂交方法检测245例新疆疑似支气管哮喘学龄前患儿(哈萨克族48例、维吾尔族68例、汉族129例)的FCER2 T2206C及LTC4S A-444C基因多态性,分析基因型频率和等位基因频率在不同民族及不同地区相同民族间是否存在差异。结果:245例患儿中,新疆地区汉族患儿与维吾尔族、哈萨克族患儿LTC4S A-444C基因型频率及等位基因频率比较差异有统计学意义(P<0.05)。新疆地区汉族患儿与维吾尔族、哈萨克族患儿FCER2 T2206C基因型频率及等位基因频率比较差异无统计学意义(P>0.05)。结论:新疆不同民族疑似支气管哮喘学龄前患儿LTC4S A-444C的基因型与等位基因的分布频率存在差异性。本研究为进一步探讨儿童支气管哮喘分子诊断方法、疾病预测及防治奠定了基础。

Abstract:
Objective: Currently there is no specific detection method and index in the diagnosis of preschool children's asthma. To analyze the difference distribution of FCER2 T2206C and LTC4S A444C in Kazak, Han and Uygur preschool children with suspected asthma of Xinjiang. To provide the early diagnosis basis of asthma for preschool children who are susceptible to asthma of different nationalities in Xinjiang. Methods: The polymorphism of FCER2 T2206C and LTC4S A444C genes in 245 preschool children with asthma in Xinjiang (including 48 Kazak, 68 Uygur and 129 Han) were detected by fluorescence staining in situ hybridization, and the differences of genotype frequency and allele frequency among different nationalities and different regions were analyzed. Results: Among 245 preschool children with suspected asthma in Xinjiang, there were significant differences in LTC4S A-444C genotype frequency and allele frequency between Han children and Uygur and Kazak children (P<0.05); There was no significant difference in FCER2 T2206C genotype and allele frequency among the three nationalities (P>0.05). Conclusion: There was statistical difference on genetic polymorphism of LTC4S A-444C between three nationalities in preschool children suspected with asthma of Xinjiang. Through this study, we can provide the basis for screening or early diagnosis of suspected asthma in preschool children of different nationalities in Xinjiang, and early intervention or treatment of asthma in children. So as to further explore the molecular diagnosis method, disease prediction and prevention and treatment of children's bronchial asthma.
