| 目的:基于系统药理学方法探索中药方剂银莱汤治疗小儿肺炎的活性成分和作用机制。方法:通过中药系统药理学数据库(TCMSP)检索银莱汤的主要活性成分,通过靶点预测和蛋白-蛋白相互作用分析银莱汤发挥药理作用的关键蛋白,进一步分析蛋白功能和富集通路,然后采用Cytoscape 3.8.2软件构建“中药-成分-靶点-通路”模型,得到关键靶点后通过动物实验进行初步验证。结果:银莱汤中34种成分与31个关键靶点具有较强的相互作用,主要涉及细胞因子及其受体的相互作用、趋化因子信号通路、白细胞介素-17信号通路、类风湿性关节炎通路、Th17细胞分化、肠道免疫反应、肿瘤坏死因子信号通路、哮喘通路和核转录因子(NF)-κB信号通路等,动物实验结果表明银莱汤对小鼠肺部炎症模型有较好的治疗效果。结论:银莱汤可以通过调控免疫反应、干预相关疾病通路等方式实现治疗小儿肺炎的作用,体现了银莱汤多成分、多靶点、多途径协同作用的特点。 |

| Active Ingredients and Mechanism of Yinlai Decoction in the Treatment of Children with Pneumonia Based on System Pharmacology |

| Objective: To probe into the active ingredients and mechanism of Yinlai decoction in the treatment of children with pneumonia based on system pharmacology. Methods: Through the traditional Chinese medicine systems pharmacology database and analysis platform (TCMSP), the active ingredients of Yinlai decoction were obtained. Through target prediction and protein-protein interaction analysis, the key proteins of Yinlai decoction were analyzed, and the protein functions and enrichment pathways were further analyzed. Cytoscape 3.8.2 software was used to construct the model of “traditional Chinese medicine medicine-ingredient-target-pathway”, and the key targets were obtained and preliminarily verified by animal experiments. Results: Totally 34 ingredients in Yinlai decoction had strong interactions with 31 key targets, mainly including the interaction of cytokines and receptors, chemokine signaling pathway, interleukin-17 signaling pathway, rheumatoid arthritis pathway, Th17 cell differentiation, intestinal immune response, tumor necrosis factor signaling pathway, asthma pathway and nuclear transcription factor (NF)-κB signaling pathway, etc. Results of animal experiments showed that Yinlai decoction had better therapeutic effects on pulmonary inflammation model in mice. Conclusion: Yinlai decoction can treat children with pneumonia by regulating the immune response and intervening the related disease pathways, reflecting the characteristics of multi-component, multi-target and multi-pathway synergistic action of Yinlai decoction. |
