| Abstract: |
| Objective: To investigate the correlation between levels of tumor necrosis factor receptor-associated factor 3 (TRAF3),
galectin-3 (Gal-3), and peroxiredoxin 1 (PRDX1) and disease severity, prognosis in children with ulcerative colitis (UC). Methods:
From Apr. 2022 to Apr. 2023, totally 176 children with UC (UC group) and 176 children undergoing routine health checkups (control
group) in our hospital were enrolled. Based on the Mayo Endoscopic Score (MES), the UC group was classified as follows: 63 cases
with MES grade Ⅰ, 71 cases with MES grade Ⅱ, and 42 cases MES grade Ⅲ. Based on 2-year follow-up outcomes, UC group was
further divided into the good prognosis group (121 cases) and poor prognosis group (55 cases). Enzyme-linked immunosorbent assay
was used to detect serum levels of TRAF3, Gal-3, and PRDX1. Pearson or Spearman correlation analysis, multivariate logistic
regression, ROC curve analysis, and the Bootstrap method were employed for internal validation. Results: Compared with the control
group, the UC group showed decreased serum TRAF3 and PRDX1 levels and elevated Gal-3 levels (P<0. 05). The levels of serum
TRAF3 and PRDX1 in children with MES grade Ⅰ to Ⅲ decreased successively, while the level of Gal-3 increased successively (P<0. 05).
The levels of TNF-α and Gal-3 in the poor prognosis group were higher than those in good prognosis group, while the levels of SOD,
TRAF3 and PRDX1 were lower in poor prognosis group (P<0. 05). Serum TRAF3, PRDX1 were negatively correlated with Gal-3, MES
grade, and TNF-α, and positively correlated with SOD. Gal-3 was positively correlated with MES grade and TNF-α, and negatively
correlated with SOD. Serum TRAF3 was positively correlated with PRDX1 (P<0. 05). TNF-α, Gal-3, SOD, TRAF3, and PRDX1 were
identified as risk factors for poor prognosis (P<0. 05). The AUC for predicting poor prognosis using the three variables in combination
was superior to that of any single predictor. The AUC after bootstrap correction was 0. 927 (95% CI 0. 883 to 0. 958); the Hosmer-
Lemeshow test yielded χ2 =6. 324, P = 0. 612. Conclusion: Children with UC have reduced serum levels of TRAF3 and PRDX1 and
elevated levels of Gal-3, all three markers are closely associated with disease activity scores and are highly valuable for predicting poor
prognosis when used in combination. |
