Abstract:
Objective: To retrieve literature on neonatal adverse drug reactions induced by linezolid (LZD) and collect relevant information, so as to provide reference for safe clinical medication. Methods: Wanfang, CNKI, Wiley Onlinelibrary and PubMed databases (the retrieval time was up to Mar. 15, 2025) were retrieved to collect clinical data of neonates with adverse drug reactions induced by LZD during hospitalization. The basic information was extracted, and the types and causes of adverse drug reactions were statistically analyzed. Results: A total of 15 literature on adverse drug reactions induced by LZD in the neonates were collected, including 553 children, with 317 males (57. 32%) and 236 females (42. 68%). Gestational ages ranged from 24 to 42 weeks, with birth weights ranging from 350 to 4,300 g and postnatal ages ranging from 1 to 47 d. Adverse drug reactions were observed in 107 cases (19. 35%). Hematological changes were the most common, reported in 81 cases, with thrombocytopenia of varying severity observed in 63 cases. Gastrointestinal symptoms were reported in 21 cases, metabolic disorders in 3 cases, and neurological lesions in 2 cases. Following the discontinuation of LZD and symptomatic supportive treatment, 2 cases with loss to follow-up, adverse drug reactions were resolved in 104 cases, except for one case of irreversible auditory neuropathy. Conclusion: The main adverse drug reaction of LZD in neonates was thrombocytopenia, which could be return to normal state after withdrawal. Hyperlactatemia and auditory nerve injury are rare. Continuous monitoring during treatment and timely ophthalmologic and hearing assessments are recommended to ensure medication safety.
