儿童特应性皮炎皮肤屏障功能相关蛋白表达情况

摘要:
目的:了解特应性皮炎患儿皮肤屏障功能相关蛋白的表达情况。方法:应用免疫组化方法检测特应性皮炎患儿和正常儿童的中间丝聚合蛋白、内披蛋白、兜甲蛋白等皮肤屏障功能相关蛋白(表皮终端分化蛋白)的表达差异,采用染色强度(0~5 分)反映相关蛋白的表达水平。结果:正常儿童皮肤的中间丝聚合蛋白、内披蛋白、兜甲蛋白的染色强度分别为(4.49±0.14)分、(4.39±0.14)分、(4.56±0.15)分,特应性皮炎患儿受累部分皮肤分别为(2.19±0.17)分、(2.29±0.11)分、(1.89±0±22)分,两组比较差异均有统计学意义(P均<0.01)。特应性皮炎患儿和正常儿童的中间丝聚合蛋白、兜甲蛋白均主要表达于颗粒层,正常儿童内披蛋白主要表达于棘细胞上层和颗粒层,特应性皮炎患儿内披蛋白主要表达于棘细胞上层。结论:特应性皮炎患儿皮肤屏障功能相关蛋白表达下降,提示存在皮肤屏障功能障碍。

The Expression of Skin Barrier Function Proteins in Pediatric Atopic Dermatitis

Abstract:
Objective: To investigate the expression of skin barrier function proteins in pediatric atopic dermatitis. Methods: Different expressions of filaggrin, involucrin and loricrin from atopic dermatitis children and normal children were detected by immunohistochemistry. The levels of associated proteins were reflected by staining intensity (0 to 5 score). Results: In atopic dermatitis children, the staining intensitys of filaggrin, involucrin and loricrin reduced significantly compared with normal children (2.19±0.17 vs 4.49±0.14, 2.29±0.11 vs 4.39±0.14, 1.89±0.22 vs 4.56±0.15, respectively, P<0.01). In atopic dermatitis children and normal children, filaggrin and loricrin were primarily expressed in the stratum granulosum of the skin, involucrin was expressed in the upper sprickle cells layer and stratum granulosum in normal children. However, involucrin was mainly expressed in the upper sprickle cells layer in atopic dermatitis children. Conclusion: The expression of skin barrier function related proteins are decreased and skin barrier function in atopic dermatitis is damaged.
