目的:探讨白细胞介素4(IL-4)和白细胞介素13(IL-13)基因多态性对哮喘患儿IgE表达量的影响。方法:采用聚合酶链式反应(PCR)观察173例哮喘患儿及56例健康儿童的IL-4和IL-13基因限制性长度片段多态性,及不同基因型患儿IgE的表达水平。结果:哮喘患儿IL-4-589位点基因多态性和等位基因与健康对照儿童比较差异无统计学意义(P>0.05)。哮喘患儿IL-13基因+1923位点基因多态性和等位基因与健康对照儿童比较差异有统计学意义(P<0.01)。IL-4基因-589和IL-13基因+1923位点T等位基因患儿IgE水平显著高于C位点患儿(P<0.01)。结论:哮喘患儿IL-13基因多态性与健康儿童不同,IL-4基因-589位点和IL-13基因+1923T等位基因是导致哮喘患儿IgE水平升高的可能原因。 |

The Influence of IL-4 Gene and IL-13 Gene on IgE Level in Asthma Children |

Abstract: |
Objective: To study the influence of interleukin-4 (IL-4) gene and interleukin-13 (IL-13) gene on IgE level in asthma children. Methods: PCR was applied to study the restriction fragment length polymorphism (RFLP) of IL-4 gene and IL-13 gene in 173 asthma children and healthy control and the IgE expression level of different genotypes. Results: The RFLP and alleles of IL-4-589 in asthma children has no significant difference between healthy control (P>0.05). The RFLP and alleles of IL-13+1923 in asthma children were significantly different between two groups (P<0.01). The IgE level in asthma children with T allele of IL-13+1923 gene and IL-4-589 were significantly higher than those in C allele (P<0.01). Conclusion: The IL-13 genotype was different from healthy control and the T genotype of IL-4-589 and IL-13+1923 may play a important role on the increase of IgE in asthma children. |
