氯胺酮对SD幼鼠海马GAP-43蛋白的影响分析

摘要:
目的:分析氯胺酮对SD幼鼠海马GAP-43蛋白表达的影响。方法:取7~10日龄的SD幼鼠36只,随机分为A、B、C三组各12只。A组注射生理盐水50 mg/kg,B组注射氯胺酮25 mg/kg,C组注射氯胺酮50 mg/kg。24 h后处死小鼠取材,分别采用免疫组化与Western Blotting检测海马GAP-43蛋白的表达情况。结果:经图像分析软件处理后得知,A组B组、C组条带灰度值分别为187.43±7.98、164.87±8.35、154.31±6.98,组间比较差异均有统计学意义(P<0.01)。A组、B组、C组GAP-43阳性细胞计数分别为(87.45±14.98)个/mm2、(66.48±12.36)个/mm2、(35.32±9.93)个/mm2,组间比较差异均有统计学意义(P<0.01)。结论:氯胺酮可明显降低SD幼鼠海马GAP-43蛋白的表达,这可能与其影响幼鼠神经系统发育有关。

Abstract:
Objective: Analysis on the effect of ketamine on the expression of GAP-43 protein in hippocampus of SD rats. Methods: 7~10 day old SD rats 36, were randomly divided into three groups. A group of 12 rats was injected with saline 50 mg/kg, group B 12 rats was injected with ketamine 25 mg/kg, group C 12 rats was injected with ketamine 50 mg/kg. 24 h after the mice were sacrificed and drawn.Immunohistochemistry and Westernblotting were used to detect the expression of hippocampal GAP-43 protein. Results: After image analysis software processing that, A group of gray stripe is 187.43±7.98, B group striped gray value of 164.87±8.35, Group C striped gray value of 154.31±6.98, the differences between groups with statistical difference (P<0.01). The A group of GAP-43 positive cell count was (87.45±14.98)/mm2, B group GAP-43 positive cell count was (66.48±12.36)/mm2, C group GAP-43 positive cell count was (35.32±9.93)/mm2, comparison between groups with statistical difference (P<0.01). Conclusion: Ketamine can significantly reduce the expression of SD rats GAP-43 protein in the hippocampus, which may be related to the impact related to the development of the nervous system in young rats.
