酶渗漉与醇渗漉提取颠茄草中莨菪碱的效果比较

摘要:
目的:应用酶渗漉法和醇渗漉法分别提取颠茄草中主要活性物质莨菪碱,考察两种溶媒提取的提取率,优选最佳提取方法。方法:用pH值为2.0、体外消化活力为24~30 U/mL的胃蛋白酶溶液为溶媒渗漉颠茄草,以80%乙醇为溶媒提取的渗漉液做对照,用高效液相色谱法测定两组提取液中莨菪碱的含量,比较两种溶媒的提取率。结果:酶渗漉法莨菪碱提取率为4.87 mg/g,醇渗漉法提取率为3.66 mg/g,酶渗漉法比醇渗漉法提取率高33.1%(P<0.05)。结论:应用酶渗漉法提取颠茄草能显著提高莨菪碱的提取率,降低成本,具有实用价值。

Abstract:
Objective: Enzyme percolation method and alcohol percolation method wererespectively utilized to extract herba belladonnae. Extraction rates of the main active substance-hyoscyamine from two methods were investigated. The best extraction method was selected. Methods: In the experimental group, the pepsin solution, pH was 2.0, in vitro digestion energy was 24~30 U/mL, was utilized as solvent to percolate herba belladonnae. In the control group, the 80% ethanol solution was utilized as solvent to percolate herba belladonnae. High performance liquid chromatography was utilized to test thecontent of hyoscyamine in extract of two groups and extraction rates from two groups were compared. Results: The extraction rate of hyoscyamine in experimental group was 3.66 mg/g, it was 33.1% higher than the number in control group which was 3.66 mg/g (P<0.05). Conclusion: Enzyme percolation method used in extracting herba belladonnae can significantly increase the extraction rate of the main active substance- hyoscyamine. It can help reducing cost and has practical value.
