云南地区哮喘患儿GLCCI1基因rs37973位点多态性分布特点

摘要:
目的:了解云南地区哮喘患儿GLCCI1基因rs37973位点的分布特点,为临床针对不同地域、种族和基因型的个体实现药物个体化治疗提供参考。方法:选取2016年11月至2018年10月在昆明市儿童医院门诊或住院部被诊断为哮喘的患儿共157例,采用荧光原位杂交法(FISH)检测哮喘患儿GLCCI1基因rs37973位点的基因型。结果:157例患儿的GLCCI1基因rs37973位点基因型分布为GG 36例(23.0%)、GA 77例(49.0%)、AA 44例(28.0%),等位基因G和A的分布频率分别为47.5%和52.5%。基因型分布与欧洲、南美洲、东南亚、东亚和部分南亚及部分北美洲地区基本一致(P>0.05),而与非洲、西亚、中美洲和部分南亚以及部分北美洲地区存在差异(P<0.05)。结论:哮喘患者GLCCI1基因rs37973位点的基因型分布可能具有地域、种族差异,临床应该针对性地给予个体化药物治疗。

Distribution Characteristics of Polymorphism of rs37973 Locus in GLCCI1 Gene in Children with Asthma in Yunnan

Abstract:
Objective: To investigate the distribution characteristics of polymorphism of rs37973 locus in GLCCI1 gene in children with asthma in Yunnan, so as to provide theoretical basis for the realization of individualized drug therapy for different regions, races and genotypes. Methods: A total of 157 children diagnosed as asthma in outpatient or inpatient department of Kunming Children’s Hospital from Nov. 2016 to Oct. 2018 were extracted. Fluorescence in situ hybridization (FISH) was used to detect genotype of rs37973 locus in GLCCI1 gene in children with asthma. Results: The polymorphism of rs37973 locus in GLCCI1 gene was 36 cases (23.0%) in GG type, 77 cases (49.0%) in GA type and 44 cases (28.0%) in AA type. The distribution frequencies of allele G and A were 47.5% and 52.5%, respectively. The genotype distribution was basically consistent with Europe, South America, Southeast Asia, East Asia and parts of South Asia and parts of North America (P>0.05), yet different from that of Africa, West Asia, Central America and parts of South Asia and parts of North America (P<0.05). Conclusion: The distributions of rs37973 locus in GLCCI1 gene in children with asthma may have regional and ethnic differences, and individualized drug therapy should be given for different regions and genotypes in clinical practice.
