在神经母细胞瘤中八聚体结合转录因子4的表达及与瘤细胞侵袭转移的关系初探

[摘要]目的:观察八聚体结合转录因子4(Oct4)在神经母细胞瘤中的表达情况,并探讨其与神经母细胞瘤转移和侵袭的关系。方法:免疫组织化学方法和RT-PCR方法检测神经母细胞瘤组织及癌旁组织中Oct4的表达情况。收集患者临床病理参数,分析Oct4表达与神经母细胞瘤临床病理参数之间的关系。采用电穿孔转染法,将Oct4 siRNA转入神经母细胞瘤SK-N-MC细胞中,通过伤口愈合实验和Boyden小室检测Oct4对神经母细胞瘤SK-N-MC细胞转移和侵袭能力的影响。结果:神经母细胞瘤组织中Oct4的表达水平高于癌旁组织(P<0.05)。Oct4的阳性表达率与患者有无淋巴结转移和分化程度密切相关(P<0.01),但与患者性别、年龄、肿瘤大小和临床病理分期均无明显关系(P>0.05);转染Oct4 siRNA可显著抑制神经母细胞瘤SK-N-MC细胞的转移和侵袭能力(P<0.01)。结论:Oct4具有促进神经母细胞瘤转移和侵袭的作用,其高表达可能与神经母细胞瘤的发生和发展密切相关。

Expression of Octamer-Binding Transcription Factor 4 in Neuroblastoma and Its Correlation with Invasion and Metastasis of Tumor Cells

Abstract:
[Abstract] Objective: To observe the expression of octamer-binding transcription factor 4 (Oct4) in neuroblastoma, and probe into its correlation with metastasis and invasion of neuroblastoma. Methods: The expression of Oct4 in neuroblastoma tissues and paracancerous tissues was detected by immunohistochemistry and real-time PCR (RT-PCR). The clinicopathological parameters of the patients were collected, and the correlation between expression of Oct4 and clinicopathological parameters of neuroblastoma was analyzed. The electroporation transfection method was used to transfer Oct4 siRNA into neuroblastoma SK-N-MC cells. Effects of Oct4 on metastasis and invasion of neuroblastoma SK-N-MC cells was detected by wound healing assay and Boyden chamber. Results: The expression level of Oct4 in neuroblastoma tissues was higher than that in paracancerous tissues (P<0.05). The positive expression rate of Oct4 was closely related to the presence or absence of lymph node metastasis and differentiation (P<0.01), but it is not significantly related to patient’s gender, age, tumor size and clinical pathological stage (P>0.05). Transfection of Oct4 siRNA can significantly inhibit the metastasis and invasion of neuroblastoma SK-N-MC cells (P<0.01). Conclusion: Oct4 can promote the metastasis and invasion of neuroblastoma, and its high expression may be closely related to the occurrence and development of neuroblastoma.
